Didanosine is an oral drug that is used for the treatment of human immunodeficiency virus (HIV), which is responsible for the acquired immunodeficiency syndrome (AIDS). During infection, the HIV virus reproduces within the body cells; as a result, the new viruses are released from the origin cell and spread to other uninfected cells. Therefore, the infection spreads to healthy cells and it replicates its reproduction. During the production of the new viruses, the HIV virus creates a new virus that contains a new DNA in every single virus. Therefore, Didanosine is converted within the body into an active form known as dideoxyadenosine triphosphate, which is a chemical similar to deoxyadenosine triphosphate, a chemical required by the HIV virus to create new DNA. Consequently, the reverse transcriptase uses the active form of Didanosine for creating the DNA; as a result, interferes with the reverse transcriptase.

Didanosine is used in the treatment of HIV in both adults and Children. Therefore, you should consult with the doctor on the dosage that you require or adhere strictly to the manufacturer's guidelines. Didanosine is found in the form of capsules (extended release) 125, 200, 250, and 400 mg with a solution of 10mg/ml. the capsules and unmixed powder must be stored at room temperature, 15 C to 30 C (59 F to 86 F). Additionally, the powder may be stored in the refrigerator at 2 C to 8 C (36 F to 46 F) for up to 30 days after it is mixed with water. The dosage of Didanosine is as follows:
- Adults with the weight of above 60kg should receive 400 mg once daily of the capsules or 200 mg twice daily of the powder. Adults with the weight of below 60kg should receive 250 mg once daily of the capsules and 125 mg twice daily of the powder.
- Children dosage must be strictly under the physician's advice.
Note that Didanosine must be administered on an empty stomach because food digestion minimizes the absorption of Didanosine by 46%.
Didanosine Side Effects
Didanosine like any other medication has its side effects that cannot be anticipated. However, you must immediately inform your doctor in case you have a history of liver or pancreas disease. The reason is that Didanosine can cause severe life-threatening effects on your pancreas or liver. Therefore, it is important that you inform your doctor in the case that you have a history of the following:
- A history of pancreatitis;
- Liver disease;
- A history of numbness or tingling in your hands or feet, including Raynaud's syndrome; or
- If you also take stavudine (Zerit)
